We studied the effects of a high fat high fructose diet (HFFD) on intermediary metabolism and the development of\u00a0<a class=\"topic-link\" title=\"Learn more about oxidative stress from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/oxidative-stress\">oxidative stress<\/a> in mouse liver and tested the ability of alpha-ketoglutarate to prevent HFFD-induced changes.<br \/>\nMale mice were fed a standard diet (10% kcal fat) or HFFD (45% kcal fat, 15% kcal fructose) with or without addition of 1% alpha-ketoglutarate (AKG) in drinking water for 8\u00a0weeks.<br \/>\nThe HFFD had no effect on body mass but activated fructolysis and\u00a0<a class=\"topic-link\" title=\"Learn more about glycolysis from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/glycolysis\">glycolysis<\/a>\u00a0and induced inflammation and oxidative stress with a concomitant increase in activity of antioxidant enzymes in the mouse liver. HFFD-fed mice also showed lower mRNA levels of\u00a0<a class=\"topic-link\" title=\"Learn more about pyruvate dehydrogenase kinase from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/pyruvate-dehydrogenase-kinase\">pyruvate dehydrogenase kinase<\/a>\u00a04 (PDK4) and slightly increased intensity of\u00a0<a class=\"topic-link\" title=\"Learn more about mitochondrial respiration from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/mitochondrial-respiration\">mitochondrial respiration<\/a>\u00a0in liver compared to mice on the standard diet. No significant effects of HFFD on transcription of\u00a0<em><a class=\"topic-link\" title=\"Learn more about PDK2 from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/pdk2\">PDK2<\/a><\/em>\u00a0and\u00a0<em>PGC1\u03b1<\/em>, a peroxisome proliferator-activated receptor co-activator-1\u03b1, or protein levels of p-AMPK, an active form of AMP-activated protein kinase, were found. The addition of AKG to HFFD decreased oxidized\u00a0<a class=\"topic-link\" title=\"Learn more about glutathione from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/glutathione\">glutathione<\/a>\u00a0levels, did not affect levels of\u00a0<a class=\"topic-link\" title=\"Learn more about lipid peroxides from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/lipid-peroxide\">lipid peroxides<\/a>\u00a0and\u00a0<em>PDK4<\/em>\u00a0transcripts but increased activities of\u00a0<a class=\"topic-link\" title=\"Learn more about hexokinase from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/hexokinase\">hexokinase<\/a>\u00a0and\u00a0<a class=\"topic-link\" title=\"Learn more about phosphofructokinase from ScienceDirect's AI-generated Topic Pages\" href=\"https:\/\/www.sciencedirect.com\/topics\/biochemistry-genetics-and-molecular-biology\/phosphofructokinase\">phosphofructokinase<\/a>\u00a0in mouse liver.<br \/>\n<\/span>[\/vc_column_text][nectar_btn size=&#8221;large&#8221; open_new_tab=&#8221;true&#8221; button_style=&#8221;see-through&#8221; button_color_2=&#8221;Accent-Color&#8221; color_override=&#8221;#2954aa&#8221; icon_family=&#8221;default_arrow&#8221; text=&#8221;Read the full article&#8221; url=&#8221;https:\/\/www.sciencedirect.com\/science\/article\/abs\/pii\/S0304416522001441&#8243;][\/vc_column][\/vc_row]\n","protected":false},"excerpt":{"rendered":"<p>Diets rich in fats and\/or carbohydrates are used to study obesity and related metabolic complications.
